What is the big difference between grumpy team member having a bad day and chronic complainer with perpetual bad mood?

It is important to intervene in order to assess the validity of the complaints to help find solutions and to coach team members on improved communication skills.

Maintaining an open door policy can help create a culture that prevents chronic complaining.

The former is normal and I'd expect to see the rest of the team supporting the team member. The latter is toxic and you are likely to see a very low level of support or patience for the behavior from other team members and/or others adopting the same bad behaviors...

In my experience chronic complainer will find problems and mistakes to be threatening, so they try to hide them most of the time. It's not easy to deal with people like this.

You might be interested to deliver special courses for your negative employee to help him out reduce some of the negativity energy and turn it to useful positive energy for healthier work environment
