The scope should be defined early, the First stage is Initiating! The contract comes before that!
In PMBOK it is in the Planning stage.

Defining scope is not a high-level requirement, So in the project charter, it can not be.
Defining scope means to know all and only work which is required to be done as part of the project. So in the planning process group scope is clearly defined utilizing Available sources of information. If any further information becomes available in future the already defined scope will be amended through the change control process.
