Wow. That is a broad question that can be answered in many different ways.

From PMI... "A management structure that standardizes the program-related governance processes and facilitates the sharing of resources, methodologies, tools, and techniques."

Can you be specific on where you are coming from and why you are asking? There is a lot of literature out there on how to setup a PMO.

It would depend on the type of PMO that you are setting up - 1) Administrative or 2) Functional. It would also depend on your reasons for implementing a PMO - 1)making optimal use of available resources, 2) ensuring PM processes are delivered in a correct, consistent, and timely manner, and 3) providing the leadership, organizational structure, regulations, standards and processes that ensure projects comply with best practices in accordance with your enterprise, mission, strategy, and business objectives. I too am involved in setting up a PMO and found this information online from PA&E Global - "The Project Management Office (PMO) Best Practices and Processes"

The best answer of your question is:
Sit back and relax write a one page problem statement
why you want to setup a PMO. This way you know the actual problem and might be setting up the PMO is not the right solution

If the problem statement clearly states that , there is really a need to setup a PMO then you have to first write down the scope of work for setting up the PMO same as you write project SRS Document
This entails, you the boundary of PMO whats IN Scope and whats out of scope and get your PMO SOW signed by one of our company executive then you have a clear roadmap for the PMO for your organization.

I would focus on the existing needs of your organization. We have a new PMO at our organization, and we were already managing one corporate program, but our role is not to house PMs to manage all projects within the organization.

What we are working on is defining the areas where our organization could use some PMO value, such as portfolio management, change control... We are taking small strides in order to ensure that our value is achieved.

Good luck!
