I would recommend that you develop a mission statement for your PMO along with key Goals and objectives. Then review, adjust and align it with your stakeholders first. That way everyone understands the purpose of the PMO and what is expected from it. PMO's have different roles and scope in different companies, so it is important to get this alignment early. I have never been in or part of a PM startup... but however, make sure you aren't a jack of all trade... Study the market and put forth your prime PM skills

Is not about to start a PM firm. Is about to start a firm directly. To do that you have to come to literature on the matter mainly those in the business administration domain.

Main hurdles to overcome are not PM specific, but generally how to start a company.

You may read books like 'sense and respond' which give you practical advice.
And get a mentor.
