Knowing all the methodologies and how to do them is important, but I think you definitely need to have some flexibility. I am in a non-profit organization and there is no way our organization can do full-on agile or scrum to the nth degree. But having the user story backlog in Visual Studio Team Services, the two week sprints, the negotiation as to what is worked on at every two week sprint meeting, and ensuring that the user gets their hands on a working piece of software as soon as possible has been instrumental in how the customer perceives their software product.
